It dimerizes with other bHLH transcription factors and activates transcription through binding to E-BOX ELEMENTS containing 5'-ACANNTGT-3' or 5'-CCANNTGG-3' sequences. Mutations in the TCF-4 gene are associated with Pitt-Hopkins Syndrome, a severe developmental disorder.
