    I am physicist by training and previously directed proteomics facilities; for over 20 years I was involved in biological mass spectrometry, from instrument development to method design to proteomics data interpretation and validation. I recently transitioned to biomedical research via a K25 Mentored Career Development Award to study the impact of prenatal exposure to toxic lipids on mitochondrial function in kidney and liver of offspring of obese mothers. This transition allowed me to learn about biology, biochemistry, cellular function and integrated physiology; data I used to supplement my coursework and enhance my learning were recently published in PlosOne: “Spaceflight activates lipotoxic pathways in mouse liver.” My K award project is focused on the role of SIRT3 in developmental programming and the potential of blunting mitochondrial dysfunction resulting from lipid toxicity and oxidative stress using PQQ, a novel antioxidant, as a potential therapeutic. My data and recent observations suggest that PQQ activates SIRT3 and protects mice from hepatic fat accumulation and suppresses pro-inflammatory and pro-fibrotic gene expression, as well as more systemic metabolic dysfunction. Our new data point toward altered glycolytic metabolism as a novel driver of developmental programming of nonalcoholic steatohepatitis, leading to our new hypothesis that maternal obesity leads to metabolic reprogramming of hepatic macrophages via HIF1a-mediated acceleration of glycolysis and induction of pro-inflammatory genes and that PQQ treatment in early life reverses the programming and provides persistent hepatic protection by altering expression of histone demethylases and deacetylases.
