    The Effects of Imatinib on Endothelial Dysfunction in Inflammatory Lung Disease
    Role Description: The proposed study employs a comprehensive and highly mechanistic approach to determine the mechanisms by which imatinib attenuates LPS-induced Acute Lung Injury (ALI), but exacerbates Ventilator-Induced Lung Injury (VILI). Our approach will utilize our established LPS-induced ALI and VILI cell culture models (LPS and 18% CS) as well as preclinical murine models (LPS and MV) to elucidate the cellular mechanisms of action and validate their clinical significance.

    Functional Characterization of ALI-Associated MYLK SNPs
    Role Description: The proposed research aims to functionally characterize ALI-associated MYLK SNPs at the cellular level and validate these findings in established preclinical models of acute lung injury (ALI) and ventilator induced lung injury (VILI). This research utilizes state-of-the-art biophysical and ultrastructural technologies including transendothelial resistance measurements (TER), confocal live cell imaging and co-localization, kymography and fluorescence resonance energy transfer (FRET). Additionally, it involves highly translational studies using preclinical ALI/VILI models that aim to verify nmMLCK inhibition as a novel therapeutic strategy in ALI.
